ジアミン/ポリアミン

化学工業にとって重要な原料であるエチレンジアミン、ポリアミンならびにその高級同族体のご紹介です。

ポリアミンのカスタム合成

エチレンジアミンのようなポリアミンおよびその高級同族体は、化学工業にとって重要な原料です。putrescine、spermidineおよびspermineのような化合物は真核生物および原核生物のいずれにおいても重要な役割を担っており、様々な生物学的機能を示します。

ほとんどの真核生物細胞はポリアミントランスポーターシステムを細胞膜上に有しており、外来性のポリアミンの取り込みを促進します。このシステムは、急速に増殖している細胞中で高活性を示し、現在開発中の多くの治療薬の標的となっています。

これらはまた翻訳の際のプログラムされたリボソームフレームシフトのプロモーターとして作用することがわかっています。細胞は、複雑かつ高度に秩序立った経路を通じて各種のポリアミンを合成します。ポリアミンは陽イオンであるためDNAに結合し、構造的に見ると決められた間隔で並んでいる正電荷を持つ化合物となっています。細胞によるポリアミンの合成が阻害されると、細胞増殖は停止するか、または非常に遅くなりますが、外因性ポリアミンの供給によリ細胞の増殖は回復します。

ポリアミンはまたNMDA受容体およびAMPA受容体を含む様々なイオンチャンネルの重要な調節因子でもあリます。これらは内部整流カリウムチャンネルを遮断し、それにより細胞エネルギーポテンシャル、すなわち細胞膜の反対側へのK+イオン勾配を維持します。加えて、ポリアミンは、ColicinE7オペロンのsos応答の発現の開始に関与し、colicinE7の取り込みに不可欠なタンパク質の発現を抑制し、その結果ストレス状態下にあるcolicinを生産しているE.coliに対して延命効果をもたらします。

ポリアミンはまた血液脳関門の透過性を高めることができます。植物の器官の老化の調節にも関与しており、植物ホルモンとして働いていると考えられています。

DNAと相互作用するスペルミジン、スペルミンとその他の生体由来ポリアミン

ポリアミンは核酸を化学分解および酵素分解に対して安定化させ、二次構造および三次構造の形成と細胞による取リ込みを促進します。

putrescineおよびspermidineのような生物由来のポリアミンは多様な生物学的プロセスに関与しています。

例えばポリアミンはDNAおよびRNAに結合することにより、核酸の機能発現において重要な役割を果たし、DNAの凝縮および凝集を生じDNAの立体構造の変化を引き起こします。

スペルミン

Spermine(FFFH) (N2,N3,N4-Tris-Fmoc-1,5, 10,14-tetraaza-quatrodecan)は、遊離のアミノ基によって分子内のFmoc保護基が切断されるため不安定です。

この反応の速度は遅いため、Spermine(FFFB:製品コードSNNlOOO)を使用し、以下の手順にしたがってBoeを除去することにより、Spermine(FFFH)をin situ で生産することができます。

80mLの1MのHCl/MeOH中に2.65g(3.1 mmoL)のN1-Boc-N2,N3,N4-tri-Fmoc-spermineを溶解させ、この混合物を25 ℃ で6時間撹拌します。

脱保護の進行はTLC(トルエン:EtOH :AcOH 40:10 :1) によリモニターすることができます。

完了後、ロータリーエバポレーターによって減圧下で溶媒を蒸発させて乾燥させます。約50mlの塩化メチレンで再溶解し、再度蒸発させて乾燥させます。

残っている粘着性のガラス状油性物質は、引き続き行われる化学反応のためにすぐに溶媒に溶解させ、翌日まで保存することなくご使用下さい。これが困難である場合は、数滴のAcOHでpH4に調整したトルエン中で生成物を+4℃で数日間保存することができます。
