クリック反応(Huisgen[3+2]付加環化反応)
アジドとアルキンは[3+2]双極子付加環化反応の1種できわめて高収率かつ高い官能基選択性で進行します。クリックケミストリーの代名詞的反応として知られ、このため俗にクリック反応とも呼ばれます。したがって本ページでもこれに倣うこととします。
ほかにどのような官能基があっても、ほぼアルキンとアジドのみが反応し単一の生成物を生じるため、様々な官能基と併用可能です。反応は不可逆的で、溶媒を選ばず、水中でも反応は進行します。化学変換プロセスの効率を示すアトムエコノミーは100%となっています。
反応は様々な官能基と併用可能なため、様々な化合物を使用することが可能で、固相合成にも用いることが可能です。
適切な銅(I)触媒の添加によって、末端アルキンの場合には100万倍近く反応が加速されることが知られています。それとともに、位置選択性も向上します。特にTBTAはCu(I)を酸化から保護し、優れた活性を示すことが知られており、最近ではルテニウムにも触媒活性があるという報告がなされています

触媒フリーのクリック反応
クリック反応や、Diels-Alder反応に代表される付加環化反応は、官能基同士を1:1で結合させる手法として有用です。イメージング、創薬、センサー開発など応用の幅は多岐に渡り、それゆえ生化学、物質工学、表面化学、高分子化学などあらゆる分野で用いられるようになっています。
2002年の報告以来、実に多くの分野で利用されているクリック反応ですが、金属触媒を使用するという点で、生体への応用の際に以下の制限があります。
▶ 高い細胞毒性
▶ 意図しないタンパク質の酸化
▶ 蛍光の阻害
したがって、金属触媒フリーの手法の開発が急がれ現在では以下の2つの手法が主流となっています。

二つの部位を通常通りの分子同士の結合に用い、余ったもう一つの部位に蛍光物質、スピンラベルで修飾するなどして、より細やかな解析が可能になります。

Cbzによって保護されたPEG誘導体は、スペーサーおよびペプチド配列の末端への修飾にお使いいただけます。Cbzは強固な保護基であり、Pd/Cなどの激しい条件でしか脱保護できないため、外部環境の変化に対しても安定した部位としても有用です。

アジド-PEG
アジドは前述のようにクリック反応で用いられるアルキンとの反応がよく知られていますが、アリルホスフィン誘導体とのStaudingerライゲーション反応も有用な選択肢の1つです。後者はP(Ph)3による還元でアミンに変換できます。アジドの最大の魅力は、通常の条件では反応性が低く安定ですが特定の条件では高い反応性を示し、高い選択性をもたらすことです。

Biotin-PEG-N3
アリルアジドは、光親和性ラベリング試薬としてよく知られています。光反応を経て窒素がかい離することにより1重項のフェニルナイトレンが生成しますが、このフェニルナイトレンは反応性が高く、周囲のC-H結合やN-H結合に対して非特異的に反応します。
PEG5000やPEG2065が持つパーフルオロフェニルアジドは、前述のフェニルナイトレンよりも安定な中間体を形成するため、非特異的な反応を抑えることが可能です。これらの化合物はエストロゲン受容体の研究やカーボンまたは有機ポリマーのコーティングのための光架橋反応剤として用いられています。
