概要
自然界に数多くの糖が存在することに対応して、これらのグリコシド結合を加水分解する多数の酵素が存在します。
Iris社では酵素基質の主成分となる糖類に発色/蛍光分子が結合した分子をご提供しております。
特異的なグリコシダーゼによる切断によって発色/蛍光分子が放出され、検出されます。
遊離する色素は可溶のものにするか不溶のものにするかは選択できます。
分光装置を用いた連続分析を行うためには系が均質な溶液であることが不可欠ですが、細胞内に局在する酵素活性や、細菌のコロニーを見るためには、不溶性で弱い拡散力を示す蛍光/発色分子が好まれます。
シグナルのバックグラウンド比をなるべく小さくするためにも、検出に用いたいパラメータが、系中に存在する他の物質による光吸収や蛍光に影響を受けないような物質を選択することが望ましいといえます。

Biotech社ではグリコシダーゼ基質のラベルとして以下のような物質をご用意しております。
表. グリコシダーゼ基質のラベル
6-ブロモ-2-ナフトール(6-Bromo-2-naphtol)および2-ナフトール(2-naphtol)は、Br-NapおよびNap基を有する基質の加水分解により放出されます。
同時に、ヘキサゾニウムパラローズアニリンなどの適切な染色試薬との結合によって対応する赤褐色のアゾ染料が形成されます。
ナフトールはまた、蛍光分析により直接検出することも可能です。
ニトロフェノールは塩基性条件下でニトロフェノラートとなり、これは405nm付近のモル吸光係数はニトロフェノールと比較して2桁以上も高い値を示すため、高い感度で脱プロトン化を検出可能です。
レゾルフィンは酸性および中性条件下で完全に水に不溶の赤色蛍光色素です。しかし、塩基性条件下では対応するフェノラートを形成し、溶解します。

発色グリコシダーゼ基質の中で最もよく知られた応用例は、おそらく青白選択によるスクリーニングに用いるX-Galでしょう(GBB1321、詳しくは2.3.9をご参照ください)。
lacZはβ-D-ガラクトシダーゼの遺伝子であり、広く使用されるレポーター遺伝子です。
活性を有するβ-D-ガラクトシダーゼを産生するクローンの同定には、この酵素により加水分解されうる基質が不可欠になります。
X-Galが加水分解されることで、着色した非拡散沈殿物が得られます。
スクリーニングに使用される細胞は、発色色素X-β-D-Gal(GBB1321)及び誘導物質IPTG(GBB1343)を含有する培地で増殖させます。
活性を有するβ-D-ガラクトシダーゼを産生するクローンのコロニーは、青色/青緑色で表示され、容易に視覚的に同定することができます。
X-Gal染色はまた、高等生物の研究でも使用され、例えば目的の遺伝子の年齢および組織特異的発現の研究などが挙げられます。
細胞溶解物中のβ-ガラクトシダーゼ活性は、4-MUbeta-D-GAL(GBB1283)由来の可溶性4-メチルウンベリフェロンの蛍光もしくはpNP-β-D-Gal(GBB1298)由来のp-ニトロフェノレートの吸光によって決定することができます。
1-O-メチルグリコシドは基質として反応性が弱く、多くの場合対応するグリコシダーゼの効果的な誘導物質として知られています。

β-D-ガラクトース-6-スルファターゼ基質
ガラクトース-6-硫酸スルファターゼ活性の有無はモルキオ病A型の診断に使用されます。

グルコース-6-リン酸
グルコース-6-リン酸(Robison-Ester、G6P)は、いわゆる「活性化グルコース」と呼ばれる細胞代謝において中心的な役割を果たしている有機分子で、グルコース-6-リン酸デヒドロゲナーゼ、グルコース-6-リン酸ムターゼまたはグルコース-6-フォスファターゼなどの多数の酵素の基質として働きます。
しかし、レポーター酵素の蛍光性基質および発色性基質としては働きません。

β-D-グルクロニダーゼ基質
一般的な大腸菌の約95%は、β-D-グルクロニダーゼ活性を有しています。このことから、発色培地は肉、乳製品、または貝類などの食品中の大腸菌の検出および菌数測定に用いられる他、泌尿器感染症の臨床検査にも用いられます。
非常に簡便で正確な飲料水中の大腸菌汚染検査として国際的に広く受け入れられており、また植物中でのgusA、大腸菌のβ-グルクロニダーゼ、遺伝子をマーカーとして含むコンストラクト発現検出のためにも使用されます。
4-メチルウンベリフェリル-β-D-グルクロニド(4-Methylumbelliferyl
beta-D-glucronide,
MUG)は、大腸菌の検出のための蛍光基質として国際的な規格(ISO,DIN)で推奨されています。

α-D-マンノシダーゼ基質
α-D-マンノシダーゼ基質は、2量体コンカナバリンAに対する糖の結合の速度論的解析への応用、蛍光消光による希釈溶液中でのレクチンの測定、および複合体の結晶構造の研究に用いられる他、血漿中のα-マンノシダーゼ活性を測定することで、α-マンノシドーシスの診断にも用いられます。

GBB1292は、α-アミラーゼの発色基質です。
キチナーゼは、生体高分子であるN-アセチルグルコサミン(主にキチン)に存在するβ-(1-4)-グリコシド結合の加水分解を触媒します。

微生物識別のための非発色分子
微生物は、それぞれ異なる代謝特性を持ち、その特性によって区別することができます。
したがって、特定の基質に対する代謝能(たとえばある基質を代謝することで成長できるかどうか)が、種の同定のために用いられます。
例えばL-アラビノースは、天然に存在するアラビノースの異性体であり、植物由来の多糖の構成成分です。
ほとんどの細菌は、微生物培養中の唯一の炭素源としてL-アラビノースを使用することを可能にする酵素類およびトランスポーター類をコードする誘導性アラビノースオペロンを含有しています。
