複数のS-S結合を有するペプチドの低コストな合成戦略

Phacmの利点は、Acmと同等の安定性と直交性を示し、PGA存在下で水によって容易に脱保護される点です。溶解性向上のためのアセトニトリル等の共溶媒の添加も可能です。水:DMSO=80:20の混合溶媒を用いることで脱保護および環化反応をワンポットで行うことができ、反応は通常およそ16時間~24時間(37℃、pH7)以内に完了します。



AcmとPhacmの互換

Acmは、Acmと同じ直交性と脱保護特性を示すシステイン保護基Phacmに置き換えることができます。Phacmには酵素PGAの存在下で水中での単純な加水分解により除去できるという利点があり、またPhacmと同時に使用することができる直交な保護基が存在します。

2つの異なる架橋が合成の目的である場合は、1%のTFAで切断することができるMmtと、切断しチオール基を生じるために最低25%のTFAを必要とするTrtが第一の選択肢として挙がってきます。多くの場合に、1%のTFAでMmtを除去した場合に反応後もかなりの量のMmtが残り、一方で、Trt基のかなりの部分が当該濃度で除去されてしまうことから、十分な純度と収率が得られないことが報告されています。

ラボスケールであればこの方法でも目的とするペプチドを必要量合成することが可能であり、目的外のぺプチドはHPLCにより除去することができるので、ある程度許容されます。しかし、このアプローチは、高純度と高収率が生産性、経済性に直結する商業スケールについては適用できません。

優れた代替技術によりこの問題を軽減することができ、場合によっては完全に排除することも可能です。Albericioらは、新しい保護基の探索を含む網羅的な研究を行って、除去に最低でも60%のTFAが必要であるdiphenylmethyl(DPM)の同定を含む非常に有用な新しい代替法を開発しました。

目標とする2つの異なるジスルフィド結合を持つペプチドの合成のための新しい組み合わせは以下の通りです:

a) Mmtを保護基として使用し、また第1のジスルフィド架橋の構築のために5%のTFAでこれを取り除くこと。これまでに使用されていたよりもわずかに高い濃度のTFAを使用することにより、全てのMmt基を保護しているチオール基から確実に分離することができます。

b) 第2のジスルフィド架橋の脱保護条件として90%のTFAとともに保護基としてDpmを使用すること。保護基と切断条件のこの新しい組み合わせは、二重架橋されたペプチドを高純度かつ高収率で得ることを可能にし、ラボスケールから商業スケールでの生産にまで利用可能です。

ジチオ- t-ブチル基(S-tBu)を用いると更に1種類の保護基を導入できますが、これは除去されるのに数時間を要するため改良の余地がありました。Albericioらは上記研究において、S-tBuと同じ直交性を持っている2,6-ジメトキシフェニル-ジチオ(S-DMP)を保護基として同定しました。これは、DTTのような求核試薬でわずか数分以内に除去することができ、自動合成機を用いた合成にも使用することができます。

合成難易度の高いペプチドの合成戦略

最新の結果に基づき、ジスルフィド架橋を複数保持する合成難易度の高いペプチドの合成戦略を以下のように発展させることが可能です:

a) 第1のジスルフィド結合のためにCys保護基としてS-DPMを使用したペプチドを設計し、続いてDTTで脱保護することができます。

b) 次に、Mmtを2番目のジスルフィド結合形成のためのチオール基の保護に使用します。切断には5%のTFAが十分に作用します。

c) 第3のジスルフィド結合形成のためにDpmによって保護し、90%のTFAでこれを脱保護します。

d) 第4のジスルフィド結合のためのチオール基はPhacmを用いて保護し、水中で穏やかに除去することにより他の全ての他の架橋を完全なまま残しつつジスルフィド結合が形成されます。

上記以外の多数の組み合わせを、個々の状況と用途に応じて適用することができます。下表では、新しい保護基を使用した方法を様々な数のジスルフィド架橋または他の誘導体化に適用する方法をご紹介します。
